[Method/ Result] Integrative analysis of transcriptome and proteome datasets from 43 previous studies revealed that mRNA and protein levels of 25 complement and complement-related components were lower in lung cancer tissues than lung normal tissues; conversely, its protein levels were higher in plasma of lung cancer patients than healthy subjects. The opposing changes may reflect the imbalance of complement system in cancer tissues and plasma of lung cancer patients. Our iTRAQ proteome study identified 31 decreased and 2 increased complement and complement-related proteins in lung cancer tissues, of which the reduced levels of 10 components were further confirmed by qRT-PCR and western blotting. Complement components are mainly synthesized and secreted into blood by liver. QSG-7701 hepatocytes co-cultured in pairs with lung cancer cells or normal bronchial epithelial (HBE) cells indicated that lung cancer cells increased QSG-7701 complement synthesis and secretion when compared to HBE cells.
